RESUMEN

Essential oils derived from plants are major ingredients in the medical and cosmetic industry. Here, we evaluated nine types of plant essential oils to identify potential candidates with antioxidant and elasticity-enhancing properties. Seven essential oils showed at least 10% radical scavenging activity at the highest concentration. Essential oils extracted from Aster glehnii, Cinnamomum cassia, Citrus unshiu, Juniperus chinensis L., and Juniperus chinensis var. sargentii significantly enhanced fibroblast viability, and oils from Cit. unshiu, J. chinensis L., and J. chinensis var. sargentii significantly increased cell proliferation and migration. Expression of extracellular matrix proteins, including collagen 1, collagen 3, and elastin, were upregulated by J. chinensis L. and J. chinensis var. sargentii oil, which also significantly enhanced the contractile activity of skin cells in a three-dimensional gel contraction assay. The results suggest that J. chinensis L. and J. chinensis var. sargentii essential oils may be potential anti-wrinkling and anti-oxidative agents for future consideration of use in the medical and cosmetic industry.

RESUMEN

Fungi act as important decomposers in the forest environment. They recycle essential nutrients, promote plant growth through mycorrhizal relationships, and act as food for small animals. Samples of 265 indigenous fungal species were collected from Mudeungsan National Park in 2020. These species were identified based on morphological, molecular, and phylogenetic analyses using the internal transcribed spacer (ITS), nuclear large subunit rRNA (LSU), and RNA polymerase II second largest subunit (rpb2) regions. Subsequently, seven species were identified as unrecorded species in Korea: Cordyceps cicadae, Dentocorticium bicolor, Hymenochaete nanospora, Physisporinus crataegi, Rigidoporus piceicola, Russula raoultii, and Scutellinia crinita. This study reveals their detailed macro- and microscopic morphological characteristics with phylogenetic trees to report them as unrecorded species in Korea.

Although Apiospora Sacc. has previously been considered a sexual morph of Arthrinium species on the basis of phylogenetic, morphological, and ecological diagnoses, a recent study delimited these as different species. Recently, 14 species, including eight new species, of marine Arthrinium have been reported from Korea. Six known species have previously been renamed as species in the genus Apiospora (A. arundinis, A. marii, A. piptatheri, A. rasikravindrae, A. sacchari, and A. saccharicola). However, the eight new species of marine Arthrinium (Ar. agari, Ar. arctoscopi, Ar. fermenti, Ar. koreanum, Ar. marinum, Ar. pusillispermum, Ar. sargassi, and Ar. taeanense) are yet to be studied, and thus the taxonomic status of these species remains to be clarified. In this study, we conducted phylogenetic analyses using the internal transcribed spacer, 28S large subunit ribosomal RNA gene, translation elongation factor 1-alpha, and beta-tubulin regions to confirm the phylogenetic position of these eight species. Based on these analyses, we re-identified the eight Arthrinium species as new combinations in Apiospora. Additionally, among the six known Apiospora species, two (A. piptatheri and A. rasikravindrae) have not previously been recorded in Korea. On the basis of morphological and molecular analyses, we report these as new species in Korea. Herein, we present scanning electron micrographs detailing the morphologies of these species, along with phylogenetic trees and detailed descriptions.

The present study investigated the anti­melanogenic activity of 10 essential oils using the B16F10 cell model. Initially, a wide range of concentrations of these essential oils were screened in order to determine their toxicity levels. The assigned non­toxic concentrations of the tested essential oils were then used to evaluate their effects on melanogenesis. The effects of the essential oils with potent anti­melanogenic activity on cell proliferation, protection against H2O2­induced cell death and the expression of certain melanogenesis­related genes, including MITF, tyrosinase, tyrosinase related protein (TRP)­1 and TRP­2 were also evaluated. The results revealed that the essential oils extracted from Citrus unshiu, Juniperus chinensis L., Zanthoxylum piperitum and Artemisia capillaris (A. capillaris) inhibited melanogenesis. However, among these four extracts, only A. capillaris extract enhanced cell proliferation, exhibited anti­H2O2 activities and decreased the expression level of TRP­1. It was demonstrated that A. capillaris extract inhibited melanin synthesis via the downregulation of the TRP­1 translational level. These essential oil extracts, particularly that of A. capillaris, may thus be used as natural anti­melanogenic agents for therapeutic purposes and in the cosmetic industry for skin whitening effects with beneficial proliferative properties. However, further studies using in vivo models are required to validate these findings and to examine the effects of these extracts on various molecular pathways.

The morphological optimization of Trichoderma harzianum was carried out using several surfactants to achieve increased cellulase production. Addition of the surfactants to the culture medium successfully modified the fungal morphology from an aggregated form to a dispersed form. Optimization of the fungal morphology increased cellulase activity up to 177%. The morphologically optimized conditions enhanced the accessibility of the fungus to substrates and thus promoted cellulase production.

Remazol brilliant blue R (RBBR) is an anthraquinone dye derived from anthracene that is decolorized by a white rot fungus, Phlebia brevispora. Interestingly, P. brevispora produces two phenomena of yellowish and pinkish colors during the degradation of RBBR. Here, we characterized the decolorization of RBBR by P. brevispora. The fungus was significantly different between the two colors via UV spectrophotometry, and the morphology of the hyphae observed in the respective color culture was also entirely different. Moreover, both of the two ligninolytic enzymes, laccase and manganese-dependent peroxidase (MnP), were remarkably stimulated in the yellowish culture at the beginning of the decolorization. It is possible that the RBBR decolorizing mechanism might be primarily related to the amount of laccase and MnP produced in the yellowish culture. Thus, the decolorized color may be rapidly estimated at initial period of incubation. In addition, GeneFishing technology revealed that two genes were differentially expressed in yellowish culture.

Gymnopus is a cosmopolitan genus of agaric fungi and consists of ~300 species. In Korea, Gymnopus represents common saprobic mushrooms, and 12 species have been reported in Korea. Several Gymnopus specimens were collected in Korea between 2008 and 2015. To identify them exactly, phylogenetic analysis was performed by means of the internal transcribed spacer region of ribosomal-DNA sequences from the collected Gymnopus specimens. Among them, G. iocephalus, G. polygrammus, and G. subnudus have not been reported in Korea. A phylogenetic tree and images are provided.

To the best of our knowledge, this is the first report on thermophilic fungi isolated in Korea. Three species of thermophiles were isolated from compost and were identified as Myriococcum thermophilum, Thermoascus aurantiacus, and Thermomyces lanuginosus. They can grow at temperatures above 50℃ and produce high levels of cellulolytic and xylanolytic enzymes at high temperatures. Notably, the considerable thermostability of the endo-glucanase produced by T. aurantiacus has made the fungus an attractive source of industrial enzymes.

Current studies of the antioxidant activity of fungal resources are mainly focused on the fruiting bodies of edible mushrooms. To access the potential of basidiomycetes in culture-state applications, extracts of solid cultures of 83 basidiomycetous fungi newly isolated from woody materials were prepared at the same concentration (10 mg/ml), and their antioxidant activities were measured using ABTS (2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid) and DPPH (2,2-diphenyl-1-picrylhydrazyl) radical-scavenging assays. Among the basidiomycetes tested, Cryptoporus volvatus, Daedalea dickinsii, Gloeophyllum abietinum, G. trabeum, Pseudomerulius curtisii and Stereum hirsutum exhibited good antioxidant activities. The EC50 value for the removal of free radicals was lowest (i.e., most effective) in the crude extract of S. sanguinolentum (19.61 g/ml), and reduced DNA damage based on a DNA nicking assay was observed for the extracts from the six basidiomycetous species above.

In the present study, wood vinegar was used to extract chromium, copper, and arsenic from chromated copper arsenate (CCA)-treated wood. The extraction efficiency for CCA elements was evaluated using various concentrations of wood vinegar, extraction temperatures, and extraction periods. The extraction efficiency for CCA elements increased with increasing the concentration of wood vinegar and the extraction conditions, resulting in maximal removal rate of copper (95.7%), followed by arsenic (92.7%) and chromium (86.3%). Since wood vinegar afforded high levels of copper extraction, its use was extended to copper-based preservative-treated wood, wherein significant extraction of copper up to 97.6% and 95.7% was obtained from alkaline copper quats (ACQ)- and copper azole (CuAz)-treated sawdust, respectively. To the best of our knowledge, this is the first study on the application of wood vinegar for the extraction of metal elements from CCA-treated wood.
